Mapping the pH-dependent activation of the TSA1 promoter. A new ser of yeast strains that contained the indicated versions of the TSA1 promoter driving the expression of phytase were created (panel A). The location of predicted transcription factor-binding sites with a p value < 2 × 10− 5 is denoted at the bottom of the cartoon. Cells were grown on YPGly and tested for phytase activity after 48 h of induction (no restoration of pH or nutrients) as indicated in Material and Methods (B and C).
