Screening exonucleases to improve HR efficiency. The positive rate (A) and CFU (B) for fusing five exonucleases to the N or C-terminus of Cas9. Exonucleases were employed to enhance HR efficiency at the FAA1 locus in the P. pastoris GS115 strain. Sixty clones from two plates were randomly picked for PCR analysis. The positive rate was from the number of correct clones divided by the total number of picked clones. The CFU numbers were calculated from the total colonies on the plates. Data are presented as means of two biologically independent samples
