Relative cortisol formation of CYP11B1 enzyme variants in microtiter plates evaluated by HPLC. Mutants H171L, Q166R/L271M and S168R/M286I/Q315E were selected in the fluorescence based screening of a random mutant library. All other mutants were created by site-directed mutagenesis. Conversion took place for 48 h as described under Material and methods with 400 μM 11-deoxycortisol and cortisol formation was evaluated by HPLC and is presented in a relative manner as mean of triplicates with respective standard deviation. Product formation of CYP11B1 Pa1 was assigned as 1.
