Specific activitiy of wtGalOx from F. graminearum or a codon-optimized variant with or without a C-terminal His 6 -tag expressed extracellularly in P. pastoris at 25°C for 120 h . After 120 h of cultivation the specific activity of GalOx in the supernatants was determined (dark grey bars). Additional 0.5 mM CuSO4 were added and the samples were incubated at 4°C and stirring overnight. After removal of excessive CuSO4 by gel filtration, the specific activity was determined again (light grey bars). Error bars represent standard deviation for 2 assay replicates.
