A NsiI restriction site comprises the ATG start codon (in pCYT) or the last two residues of SPUsp (pSEC) and allows a simple and one-step cloning of the cassettes corresponding to the mature proteins for cytoplasmic production (pCYT) or secretion (pSEC).
