Three-step pipeline combining dMSCC and robustness analysis. The pipeline comprised three steps: data acquisition, image analysis, and data analysis. In the first step, dMSCC was performed on S. cerevisiae subjected to feast-starvation oscillations (Additional file 1: Video S1). Raw image data from live-cell imaging were pre-processed in Fiji, segmented with StarDist-2D, and single cells were tracked using TrackMate. Shape descriptors and fluorescence values were extracted for the segmented cells. Performance and robustness of the collected data were analysed in R
