Creatinine is converted by three enzymes in sequential cascade: creatininase, creatinase, and sarcosine oxidase. First, creatininase catalyzes creatinine to creatine; creatinase catalyzes hydrolysis of creatine in the second step; sarcosine oxidase catalyzes sarcosine to detectable hydrogen peroxide (H2O2) in the third step.
