From: Disruption of rimP-SC, encoding a ribosome assembly cofactor, markedly enhances the production of several antibiotics in Streptomyces coelicolor

Figure 2

Effects of rimP-SC disruption on biomass and production of actinorhodin and calcium-dependent antibiotics in S. coelicolor . (A) Growth curve of S. coelicolor M145, rimP-SCDM and rimP-SCDMC. Biomass was calculated by mycelium wet weight in GYM plate. (B) Production of actinorhodin of S. coelicolor M145, rimP-SCDM and rimP-SCDMC in GYM medium. Cell cultures (50 mg) at each time point were treated with KOH (final concentration, 1 N) and the OD640 corresponding to 10 mg of mycelium was determined. (C) Bioassay of CDA in M145 and rimP-SCDM grown in DNA medium. (D) Transcriptional analysis of act II-ORF4 by real-time RT-PCR. The transcriptional level of act II-ORF4 was detected at 24, 48, 72, 96, 120 and 144 h in M145 and rimP-SCDM grown in GYM medium. (E) Transcriptional analysis of cdaR by real-time RT-PCR. The transcriptional level of cdaR was detected at 24, 48 and 72 h in M145 and rimP-SCDM grown in DNA medium. Data were presented as the averages of the results of three independent experiments in triplicate. Error bars showed standard deviations.
