A mutant plasmid improves the growth and viability of cells expressing HtdR. The growth of cells harboring pHtdR200 or pHtdR400 was examined by streaking (A) or spot assays (B) on LB agar plates supplemented with 50 μg/mL kanamycin, 100 μM of all-trans retinal, and 0.2% L-arabinose. (C) Wild type (tig+, closed symbols) and TF-deficient cells (Δtig, open symbols) harboring the indicated plasmids were grown in LB-kanamycin at 37°C and HtdR synthesis was induced by addition of 0.2% L-arabinose in mid-exponential phase (arrow).
