Clogging of ER translocons by posttranslational translocation of BTL2. This experiment was performed as in Fig. 4b, except that the secreted protein was BTL2. The abbreviations for the constructs are as in Fig. 2. Fluorescence images of ER-targeted GFP-HDEL are shown. In the “Control” sample, no BTL2 construct was expressed, and GFP-HDEL exhibited a typical ER pattern. For the BTL2 constructs with the α-factor signal sequence, most of the GFP-HDEL remained in the cytosol. For the BTL2 constructs with the Ost1 signal sequence, GFP-HDEL exhibited a typical ER pattern. Scale bar, 2 μm
