Dose-dependent inhibition of surface-displayed CK2 measured by the CE-assay. E. coli BL21(DE3) pCK2α pCK2β was incubated with different concentrations of a TBB (100 pM–10 µM) or b silmitasertib (10 pM–1 µM) for 10 min before the reaction was started. CK2 activity was determined by capillary electrophoretic quantification of product rate. The fractional inhibition values were calculated in respect to an uninhibited sample and are given as mean values (±standard deviation, n = 3). The IC₅₀ values were obtained by fitting the dose-dependent inhibition data to a sigmoidal four-parameter logistic equation.
