Addition of the artificial signal sequence M 16 to hLIF increased secretory production in K. marxianus . a) Expression of constructs containing wild type signal sequence did not show any reaction in an ELISA assay (open circle), but the supernatants from constructs containing M16-hLIF:FLAG showed a positive reaction (closed diamonds). b) Western blotting of the host strain (RAK3908; Host), a wild type hLIF:FLAG transformant (RAK10252; Wild type), and an M16-hLIF:FLAG transformant (RAK11616; M16) indicated that only the M16-hLIF:FLAG protein was secreted.
