From: High-level production of animal-free recombinant transferrin from Saccharomyces cerevisiae
Relative productivity of alternative leaders. rTf (N413Q, N611Q) was expressed using three different leader sequences, mHSA/MFα1, mHSA and invertase, using 2 μm expression plasmids containing the PDI1 gene. Plasmids pDB2711, pDB2929 and pDB3557 were transformed into the yeast strain DYB7. The transformed yeast were inoculated at OD600 = 0.1 into BMMD SFC and grown for five days, after which supernatant concentrations were determined by HPLC. Error bars indicate standard deviations (n = 4).
