The S. cerevisiae LEU2 selectable marker (yellow shaded) and transferrin gene (green/black/red) are integrated between the STB locus and the origin of replication. Bacterial sequences flanked by two repeat sequences containing the recognition target for FLP-recombinase (blue) are integrated into an inverted repeat (black triangles) causing them to be eliminated in yeast. The PDI1 gene is inserted downstream of the REP2 gene.
