From: Steroid conversion with CYP106A2 – production of pharmaceutically interesting DHEA metabolites
P450 content in the used B. megaterium strains. CO-difference spectra recorded after 24 hours cultivation of B. megaterium ATCC13368 (wt), cyp106a2-knockout strain (k.o. strain), and MS941_pSMF2.1CAA (CAA) in crude lysates. Cells were incubated at 30°C and 150 rpm in broth, which was supplemented with 35 μg/ml tetracycline for the cultivation of B. megaterium ATCC13368 cyp106a2-knockout and B. megaterium MS941 cells. The protein expression in MS941_pSMF2.1CAA cells was induced by adding 0.25% xylose to the culture at OD600 of 0.4 and subsequent incubation for 24 hours.
