Separate component-stabilization system in Streptomyces strategy. 1. S. lividans ΔTA obtention. 2. Transformation with the temperature-sensitive plasmid carrying the antitoxin (pGM160-YefMslTS). 3. Integration of the yoeBsl gene into the chromosome of the bacteria with plasmid pKC796-Tox. 4. Transformation with the expression plasmid (pNAnti-Prot) and removal of the temperature-sensitive plasmid. When the expression plasmid was lost toxin production produced cell death.
