Optimization of expression of His ₆ -NS3 [A] and NS5A-T-His ₆ [B] in E. coli strain BL21(DE3) at various temperatures and purification by Nickel affinity chromatography following gel filtration in case NS5A-T-His ₆ [B]. SDS-polyacrylamide gels stained with Commassie brilliant blue, showed wash fraction (lane 1), purified protein fractions eluted from Ni-NTA column using 250 mM imidazole (lane 2&3) and purified fraction of NS5A-T-His₆ by gel filtration as described in Methods section. The mobilities of marker proteins of known molecular mass are shown on the left.
