Improvement of SM4 medium composition using an L8 Plackett-Burman design matrix. Shake flask cultures were carried out with 25 ml of culture medium SM4 at 28°C and 180 rpm. FeCl3, vitamins solution enriched with myo-inositol and thiamin, trace-elements solution, ammonium sulfate and glutamate are used as described in Table 3. After Western blot analysis, the effect of each component on hIFN α2b production was analyzed by Modde 6.0 software. Results are mean values of three independent experiments.
